Det har gått tre år sedan Sentinel Primes förräderi och decepticons ödeläggelse av Chicago. Trots att autobots hindrat att jorden ödelagts räknas de redan som överflödiga av världens regeringar. Det är när bilmecken Cade upptäcker att långtradaren han ska slakta för delar är en autobot, den legendariske Optimus Prime, som det blir uppenbart att det finns krafter som gör allt för att autobots inte ska återaktiveras. Samtidigt uppmärksammas hela världen på ett nytt hot. Något världen aldrig skådat förr.

När stora monster stiger upp från haven inleds ett krig som kostar miljontals liv. För att bekämpa dessa Kaijus, skapar man jättelika robotar, Jaegers, som styrs av två piloter. Men även dessa har problem att ta sig an den hänsynslösa fienden. På gränsen till ett förödande nederlag vänder man sig till två osannolika hjältar. En före detta pilot och en ung tjej är plötsligt mänsklighetens sista hopp inför den stundande apokalypsen.

När unga Susan Murphy, på sin bröllopsdag, oväntat mosas av en meteor full av geggamoja från yttre rymden, växer hon märkligt nog till en längd av 15 meter och 21 centimeter. Militären rycker in, och hon tas tillfånga och förs till en hemlig statlig anläggning. Där döps hon om till Megarella och spärras in tillsammans med en massa andra märkliga monster.

En enorm järnjätte har kommit från mörkaste rymden, kraschlandat på jorden och följt med den 11-årige Hogarth Hughes hem. Nu har den unge Hogarth en stor kompis, men han har ett ännu större problem. Hur behåller man en så stor hemlighet som är 15 meter hög? Speciellt som en paranoid agent är ute efter utomjordingen och vill förstöra järnjätten - och med sig har han hela den Amerikanska armén.

Det råder anarki på USA:s gator och de kriminella har till och med nästlat sig in i företagens styrelserum. Han är programmerad att utföra det smutsiga jobbet kallt, beräknande och effektivt och han verkar till en början under den skottsäkra oövervinnelig. Men under ytan klappar ett mänskligt hjärta som brinner för rättvisa.

Keanu Reeves spelar den utomjordiska varelsen Klaatu som genom sitt besök på jorden hotar hela mänsklighetens framtid. Desperat försöker regeringar och vetenskapsmän förstå mysteriet kring Klaatus uppenbarelse. En kvinna får tillsammans med sin styvson nyckelroller i Klaatus plan och blir nu mänsklighetens enda hopp att räddas från total utplåning...

Då Decepticons såg ut att förlora kriget mot Autobots skickade de ett skepp mot jorden med ett innehåll som skulle rädda deras ras. De amerikanska astronauter som upptäckte det kraschade skeppet på månen tystade ner händelsen i vad de trodde var mänsklighetens intresse. Men när hemligheten läcker ut vaknar den slumrande fienden och kriget mellan Autobots och Decepticons kan plötsligt få en helt annan utgång.
