Veliäisen Marge-tädin leijuessa yötaivaalla Harry suuntaa kohti kolmatta vuottaan Tylypahkassa. Tuskallinen kyyti Poimittaislinjalla vie hänet Viistokujalle missä hän saa kuulla, että karannut vanki, Sirius Musta, jahtaa häntä. Näin alkaa uusi lukukausi. Tervetuloa Hotel Transylvaniaan, Draculan ylelliseen viiden tähden lomahotelliin, jossa hirviöt perheineen voivat vaihtaa vapaalle suojassa ihmisten katseilta. Eräänä erityisenä viikonloppuna Dracula on kutsunut kaikki parhaat ystävänsä – muun muassa Frankensteinin vaimoineen, muumion, näkymättömän miehen ja ihmissusiperheen – juhlistamaan rakkaan tyttärensä Mavisin 118. syntymäpäivää. Draculalle näiden legendaaristen hirviöiden kestitseminen ei ole ongelma eikä mikään, mutta viikonloppu saa yllätyskäänteen, kun eräs tavallinen poika tupsahtaa hotellille ja muuttaa kaiken!

Tapahtumat sijoittuvat vuoteen 1926, jolloin Lisko Scamander on juuri saanut päätökseen maailmanlaajuisen tutkimusmatkansa löytääkseen ja dokumentoidakseen erikoislaatuisen joukon taikaolentoja. Ennen kotiinpaluutaan Lisko tekee välipysähdyksen New Yorkissa. Lyhyeksi suunniteltu pysähdys kuitenkin pitkittyy, kun kuvioihin astuvat Jacob-niminen ei-taik (amerikkalainen nimitys jästille), kadonnut taikasalkku, sekä Liskon karanneet ihmeotukset, jotka tapaavat aiheuttaa harmia niin velhoille kuin ei-taikeille.

Ensimmäisen elokuvan lopussa Amerikan Yhdysvaltain taikakongressi vangitsi voimakkaan pimeyden velhon Gellert Grindelwaldin Lisko Scamanderin avulla. Toteuttaakseen uhkauksensa Grindelwald pakeni ja ryhtyi kokoamaan kannattajajoukkoa, josta valtaosa ei osaa aavistaakaan hänen todellisia aikeitaan nostaa valtaan puhdasveriset velhot hallitsemaan kaikkia ei-taikovia. Estääkseen Grindelwaldin suunnitelmat Albus Dumbledore värvää avukseen entisen oppilaansa Lisko Scamanderin, joka suostuu auttamaan tietämättömänä uhkaavista vaaroista. Rakkauden ja uskollisuuden rajat joutuvat koetukselle jopa lähimpien ystävien ja perheen keskuudessa, kun velhomaailma jakautuu yhä vahvemmin kahtia.

Balthazar Blake, nykypäivän Manhattanilla asuva mestarivelho, yrittää puolustaa kaupunkia arkkivihollistaan Maxim Horvathia vastaan. Balthazar ei kuitenkaan pysty tehtävään yksin, joten hän värvää oppilaakseen Dave Stutlerin, ihan tavallisenoloisen nuorukaisen, jossa on kuitenkin piilevää potentiaalia. The Pillar of Cephiro has the sole responsibility of keeping Cephiro alive and in balance with her prayers. Without Princess Emeraude, Cephiro would fall into ruin. Hikaru, Umi, and Fuu must fight off Zagato's henchman and find the Rune Gods if they ever want to get back home. They soon learn that friendship and loyalty are the only things they can rely on in the crumbling Cephiro.
